茯苓多糖散质量控制研究OA

茯苓多糖是茯苓多糖散调节免疫功能的主要活性成分.为准确控制该制剂质量,应用分光光度法中苯酚-硫酸法测定茯苓多糖散中茯苓多糖的含量,分别从溶剂、测定波长、反应温度、反应时间、5%苯酚溶液加入量、硫酸加入量6个方面进行优化,筛选出最佳试验条件,并进一步开展线性关系、精密度、稳定性、重复性、加样回收率等方法学验证.结果显示,测定茯苓多糖最优试验条件为溶剂采用50%硫酸溶液,测定波长490 nm,25℃水浴条件反应40 min,5%苯酚加入量为1.8 mL,硫酸加入量为6.0 mL;D-无水葡萄糖在10.19~61.14 μg/mL范围内线性关系良好(R2=0.999 4,n=6),平均回收率为99.31%.本法灵敏度高,精密度、稳定性、重复性良好,且准确可靠,可作为茯苓多糖散多糖含量的控制方法.

Pachymaran is the main active component of Pachymaran powder to regulate immune function.The content of pachymaran in Pachymaran powder was determined with phenol-sulfuric acid method by spectrophotometry.The optimum test conditions were selected from six aspects:solvent,wavelength,reaction temperature,reaction time,addition amount of 5%phenol solution and addition amount of sulfuric acid.The linear relationship,precision,stability,repeatability and sample recovery method were further verified.The results showed that the optimal test conditions for the determination of Pachymaria polysaccharide were 50%sulfuric acid solution in solvent,the determination wavelength was 490 nm,the reaction time was 40 minutes at 25℃ water bath,the addition amount of 5%phenol was 1.8 mL,and the addition amount of sulfuric acid was 6.0 mL.D-anhydrous glucose had good linear relationship in the range of 10.19 μg/mL to 61.14 μg/mL(R2=0.999 4,n=6),and the average recovery was 99.31%.The method has the advantages of high sensitivity,precision,stability,good repeatability,accuracy and reliability,and can be used as a control method for polysaccharide content of Pachymaran powder.
