烟草赤星病拮抗菌YZ01发酵条件优化及抑菌活性物质初探OA北大核心CSTPCD

由链格孢菌 Alternaria alternata 引起的赤星病是烟草上的主要叶部病害之一,常常造成巨大的经济损失.对烟草赤星病菌具有显著拮抗活性的菌株 YZ01 进行鉴定、活性物质探究及发酵条件优化,可为烟草赤星病的防治提供菌种资源,为指导其实际生产应用奠定基础.通过形态学、生理生化特征及 16S rRNA和gyrA基因序列系统发育分析,鉴定菌株YZ01 为贝莱斯芽胞杆菌Bacillus velezensis.无菌滤液抑菌试验发现贝莱斯芽胞杆菌 YZ0110%无菌滤液对烟草赤星病菌 Alternaria alternata 的抑菌率为86.56%,对苹果腐烂病菌Valsa mali、辣椒疫霉病菌Phytophthora capsici、番茄灰霉病菌Botrytis cinerea、苹果炭疽叶枯病菌Colletotrichum fructicola的抑菌率为 100.00%.进一步研究发现,贝莱斯芽胞杆菌YZ01发酵液可使烟草赤星病菌菌丝细胞结构受损,导致其原生质体外渗,从而抑制菌丝生长,其发酵液抑菌活性成分主要为脂肽及蛋白类物质.单因素试验法和正交实验优化菌株 YZ01 的最佳发酵培养基配方为葡萄糖 0.50%、胰蛋白胨 1.50%和磷酸氢二钾 1.50%.发酵液稳定性测定发现,其抑菌活性受温度、pH和紫外处理的影响较小.综上所述,该结果为烟草赤星病的生物防治提供了菌种资源,也为后续田间应用奠定了基础.

Tobacco brown spot,caused by Alternaria alternata,is a prominent leaf disease in tobacco cultivation,often resulting in notable economic losses.Identifying the strain YZ01 with significant antagonistic activity against Alternaria alternata,exploring its active substances,and optimizing its fermentation conditions can provide bacterial resources to control tobacco brown spot disease and guide its practical production and application.Strain YZ01 was identified as Bacillus velezensis through a comprehensive analysis of morphological,physiological,biochemical characteristics,and sequencing of 16S rRNA and housekeeping gene gyrA.Culture filtrate of YZ01 could significantly inhibit various fungi,with the inhibition rate of 86.56%on Alternaria alternata and 100.00%on Valsa mali,Phytophthora capsici,Botrytis cinerea,and Colletotrichum fructicola.Lipopeptides and proteins were identified as the main antimicrobial active components in the culture filtrate.Subsequent investigation revealed that the fermentation broth of YZ01 disrupted the hyphae cell structure of A.alternata through the action of these active components,leading to protoplast exosmosis and inhibition of mycelial growth.Through single factor and orthogonal design experiments,the optimal fermentation conditions for producing antimicrobial active components were determined.The optimal fermentation medium formula for strain YZ01 consisted of 1.50%dipotassium hydrogen phosphate,1.50%tryptone,and 0.50%glucose.The stability test of the fermentation broth showed that its activity was minimally influenced by temperature,pH,or UV treatment.These findings offer important insights for the biological control of tobacco brown spot disease and establish a basis for future field application.
