桔梗皂苷D对大鼠力竭运动性肾损伤的保护作用研究OA北大核心CSTPCD

Protective effect of platycodon platycodon D on renal injury induced by exhaustive exercise in rats

目的 分析桔梗皂苷D对大鼠力竭运动性肾损伤的保护作用.方法 用跑台训练建立力竭运动性肾损伤模型.将50只SD大鼠分为对照组(灌胃等量0.9%NaCl)、模型组(力竭运动模型)和低、中、高剂量实验组(分别灌胃25、50、100 mg·kg-1桔梗皂苷D).用试剂盒检测肾功能[尿素氮(BUN)、血肌酸酐(Scr)]、肾损伤标志物[中性粒细胞明胶酶相关脂质运载蛋白(NGAL)、肾损伤分子-1(KIM-1)]水平;用蛋白质印迹法检测核苷酸寡聚化结构域样受体家族3(NLRP3)、凋亡相关斑点样蛋白(ASC)、胱天蛋白酶-1(Caspase-1)蛋白表达.结果 对照组、模型组和低、中、高剂量实验组BUN水平分别为(5.52±0.49)、(9.98±0.85)、(8.30±0.65)、(7.12±0.47)和(5.98±0.63)mmol·L-1,Scr水平分别为(1.69±0.24)、(5.69±0.67)、(4.71±0.47)、(3.55±0.25)和(2.67±0.24)ng·mL-1,NGAL 水平分别为(207.58±28.56)、(613.42±63.87)、(521.52±55.35)、(419.89±40.85)和(278.13±26.48)pg·mL-1,KIM-1 水平分别为(12.11±1.52)、(80.42±9.56)、(69.03±7.58)、(47.32±6.67)和(24.67±3.60)ng·L-1,NLRP3 蛋白表达水平分别为0.43±0.06、1.35±0.11、1.03±0.10、0.78±0.09 和 0.53±0.06,ASC 蛋白表达水平分别为 0.25±0.03、0.74±0.06、0.60±0.06、0.44±0.05 和 0.31±0.04,Caspase-1 蛋白表达水平分别为 0.31±0.04、0.83±0.07、0.69±0.06、0.53±0.05 和 0.42±0.0 4.模型组上述指标与对照组比较,在统计学上差异均有统计学意义(均P<0.01);低、中、高剂量实验组上述指标与模型组比较,在统计学上差异均有统计学意义(均P<0.01).结论 桔梗皂苷D可通过抑制NLRP3炎性小体激活进而减轻大鼠力竭运动性肾损伤.

Objective To analyze the protective effect of platycodon platycodon D on renal injury induced by exercise exhaustion in rats.Methods The model of kidney injury induced by exhaustive exercise was established by treadmill training.Fifty SD rats were divided into control group(0.9%NaCl),model group(exhaustive exercise model),experimental-L,-M,-H groups(25,50,100 mg·kg-1 platycodon D).Renal function[blood urea nitrogen(BUN),serum creatinine(Scr)],renal injury markers[neutrophil gelatinase associated lipocalin(NGAL),kidney injury molecule-1(KIM-1)]were detected by the kit;nucleotide oligomeric domain-like receptor family 3(NLRP3),apoptosis-associated speck-like protein containing a card(ASC),cysteine aspartic acid specific protease-1(Caspase-1)protein expressions were detected by Western blot.Results BUN levels in control group,model group,experimental-L,-M,-H groups were(5.52±0.49),(9.98±0.85),(8.30±0.65),(7.12±0.47)and(5.98±0.63)mmol·L-1;Scr were(1.69±0.24),(5.69±0.67),(4.71±0.47),(3.55±0.25)and(2.67±0.24)ng·mL-1;NGAL were(207.58±28.56),(613.42±63.87),(521.52±55.35),(419.89±40.85)and(278.13±26.48)pg·mL-1;KIM-1 were(12.11±1.52),(80.42±9.56),(69.03±7.58),(47.32±6.67)and(24.67±3.60)ng·L-1;NLRP3 protein expression were 0.43±0.06,1.35±0.11,1.03±0.10,0.78±0.09 and 0.53±0.06;ASC protein expression were 0.25±0.03,0.74±0.06,0.60±0.06,0.44±0.05 and 0.31±0.04;Caspase-1 protein expression were 0.31±0.04,0.83±0.07,0.69±0.06,0.53±0.05 and 0.42±0.04.Compared with the control group,the above indexes in the model group were statistically significant(all P<0.01);compared with the model group,the above indexes of the experimental-L,-M,-H groups were statistically significant(all P<0.01).Conclusion Platycodon D can inhibit the activation of NLRP3 inflammasome to alleviate the renal injury induced by exhaustive exercise in rats.
