奶牛子宫内膜炎是造成乳品行业巨大经济损失的主要疾病之一,从传统中草药连翘中提取的连翘酯苷A具有广泛的生物活性.本研究旨在阐明连翘酯苷A对大肠杆菌内毒素(LPS)介导的奶牛子宫内膜上皮细胞(bEECs)炎性损伤的抑制作用,并在体外阐明其可能的分子机制.研究结果表明,在LPS诱导的bEECs炎性损伤模型中,连翘酯苷A显著抑制了 bEECs的损伤作用;qPCR结果表明,连翘酯苷A能显著抑制促炎细胞因子(TNF-α、IL-6和IL-1β)的释放,并具有剂量依赖性;免疫荧光与免疫印迹结果证实,连翘酯苷A显著抑制高迁移率蛋白1(HMGB1)的表达,进而阻遏p65蛋白的磷酸化.本研究表明,连翘酯苷A通过负调控HMGB1的表达,介导NF-κB信号通路对LPS诱导的bEECs炎性损伤具有保护作用.
Endometritis in dairy cows is one of the major diseases causing huge economic losses in the dairy industry.Recent studies have shown that forsythoside A,extracted from the traditional Chinese herb forsythia,has a wide range of biological activities.The aim of this study was to elucidate the inhibitory effect of forsythoside A on Escherichia coli endotoxin mediated inflammatory damage in bo-vine endometrial epithelial cells(bEECs)as well as the possible molecular mechanisms in vitro.The results showed that conotoxin sig-nificantly attenuated the damage effect of bEECs in a model of LPS-induced inflammatory injury in bEECs.Results of qPCR showed that forsythoside A significantly inhibited the release of pro-inflammatory cytokines(TNF-α,IL-6 and IL-1β)in a dose-dependent manner.Results from immunofluorescence and immunoblotting confirmed that coniferin significantly inhibited the expression of high mobility protein(HMGB1),which in turn blocked the activation of nuclear factor NF-κB.In conclusion,the findings suggest that forsythoside A protects against LPS-induced inflammatory injury in bEECs by negatively regulating HMGB1 expression,mediated by the NF-κB pathway.
