茉莉酸甲酯调控牛大力有效成分积累及相关基因表达OACSTPCD

Effects of Methyl Jasmonate on the Accumulation of Active Components and Related Gene Expressions in Callerya speciosa

目的:研究外源茉莉酸甲酯(methyl Jasmonate,MeJA)对牛大力有效成分含量的影响,探究其对异黄酮生物合成以及茉莉酸(jasmonate,JA)信号通路关键酶基因的调控模式.方法:以牛大力幼苗为材料,采用HPLC法分析MeJA对有效成分含量的影响,利用Illumina HiSeq平台进行转录组测序,并利用qRT-PCR技术验证相关基因的表达量.结果:外源MeJA诱导提高了牛大力有效成分芒柄花素与高丽槐素的含量.基于牛大力参考基因组数据库,鉴定出20 727 个差异基因(DEGs),其中9 091 个为上调基因,主要富集在植物激素信号转导、淀粉和蔗糖代谢、异黄酮生物合成等代谢通路,qRT-PCR结果验证了转录组测序结果准确可靠.外源MeJA激活了部分异黄酮生物合成和JA信号通路关键酶基因的表达,其中异黄酮生物合成途径基因 CHS、HIDs、I3'Hs、VR、PTR、CYP81E7、7IOMT、PTS等呈上调表达;JA信号通路基因LOXs、AOSs、OPR、ACXs、JAZs和MYCs等呈上调表达.结论:牛大力在响应外源MeJA诱导的过程中可激活CHS、HID-1、VR等异黄酮生物合成途径和LOX、MYC等JA信号通路关键酶基因,从而正向调控其有效成分芒柄花素与高丽槐素含量.

Objective:To study the effect of methyl jasmonate(MeJA)on the content of active components in Callerya speciosa,and explore the transcriptional regulation mode of MeJA on key enzyme genes in isoflavonoid biosynthesis and jasmonate(JA)signaling pathway.Methods:Using the three-month seedlings of Callerya speciosa as materials,the effect of MeJA on the contents of active compo-nents was analyzed by HPLC,transcriptome sequencing was performed by Illumina HiSeq platform,and the expressions of related genes were verified by qRT-PCR.Results:Exogenous MeJA increased the contents of index components in Callerya speciose,including form-ononetin and maackiain.Based on the transcriptome data of Callerya speciosa,a total of 20 727 differentially expressed genes(CDEGs)were found,of which 9 091 were up-regulated.KEGG enrichment analysis showed that DEGs were significantly enriched in plant hor-mone signal transduction,starch and sucrose metabolism,isoflavonoid biosynthesis etc.The qRT-PCR analysis confirmed the validity of the transcriptome data,indicating that the transcriptome data were reliable and accurate.Exogenous MeJA activated some key enzyme genes in isoflavonoid biosynthesis and JA signal pathway in Callerya speciosa.Among them,the flavonoid biosynthetic genes CHS,HIDs,I3'Hs,VR,PTR,CYP81E7,7IOMT and PTS and the genes involved in the JA signaling pathway,such as LOXs、AOSs、OPR、ACXs、JAZs and MYCs,were up-regulated.Conclusion:The contents of formononetin and maackiain are significantly increased through achieving by enhancing the expressions of LOX and MYC involved in JA signaling pathway and isoflavonoid biosynthetic genes CHS,HID-1 and VR response to exogenous MeJA in Callerya speciosa.
