磷酸肌醇3 激酶(phosphoinositol 3 kinase,PI3K)/丝氨酸/苏氨酸蛋白激酶B(protein kinase B,PKB,又称AKT)/哺乳动物雷帕酶素靶蛋白(mammalian target of rapamycin,mTOR)信号通路与腹膜间皮细胞损伤、腹膜间皮细胞上皮-间充质转化密切相关.中医药基于PI3K/AKT/mTOR信号通路能有效抑制腹膜血管生成、腹膜间皮细胞发生间充质转换以及降低壁层腹膜厚度、改善超滤衰竭,从而延缓腹膜纤维化进程.基于中医药干预PI3K/AKT/mTOR信号通路影响腹膜纤维化发生发展的相关研究,发现PI3K蛋白、AKT蛋白、pAKT蛋白、mTOR蛋白为中医药靶向PI3K/AKT/mTOR信号通路防治腹膜纤维化的主要靶点,并分析抑制新生血管生成和腹膜间皮细胞上皮-间充质转化为中医药靶向PI3K/AKT/mTOR信号通路防治腹膜纤维化的主要机理.
The phosphoinositol 3-kinase(PI3K)/serine/threonine protein kinase B(PKB,also known as AKT)/mammalian rapamy-cin target protein(mTOR)signaling pathway is closely related to peritoneal mesothelial cell injury and epithelial mesenchymal transi-tion of peritoneal mesothelial cells.Traditional Chinese medicine,based on the PI3K/AKT/mTOR signaling pathway,can effectively in-hibit peritoneal angiogenesis,mesenchymal transition of peritoneal mesothelial cells,reduce parietal peritoneal thickness,improve ultra-filtration failure,and thus delay the process of peritoneal fibrosis.This article explores the effects of traditional Chinese medicine inter-vention in the PI3K/AKT/mTOR signaling pathway on the occurrence and development of peritoneal fibrosis.It is found that PI3K pro-tein,AKT protein,pAKT protein and mTOR protein are the main targets of traditional Chinese medicine targeting the PI3K/AKT/mTOR signaling pathway to prevent and treat peritoneal fibrosis.Besides,the main mechanism of inhibiting neovascularization and epithelial mesenchymal transition of peritoneal mesothelial cells is analyzed as a traditional Chinese medicine targeted PI3K/AKT/mTOR signa-ling pathway for preventing and treating peritoneal fibrosis.
